Two factors that likely contribute to the error in collagen-to-elastin ratio are the limited scope of the SEM image sampling, and the fact that collagen and elastin are not the only components contributing to the near-IR spectra. The entire aortic sample contributes to the near-IR spectra. Substantial lipid signals are observed in the spectra as well as signals from collagen and elastin.
SEM sampling is limited to three SEM images from the inner and outer membranes and a single cross section of each vessel segment.
